RBR questions

? Wildwater et al, Cell 2005

 1)      After
showing the reduced protein expression via western blot (fig.1K), couldn't the
authors have fused GUS or GFP gene to the RBR gene and its native promoter to
visualize localized protein expression, (or was that already done previously)?

2)      Regarding
fig 2M and N, on page 1338 they say that ?Our data suggest that, in both
columella and LRC/epidermis, excessive stem cells accumulate which eventually
differentiate; mature root cap cells gradually reach smaller final sizes.? Does
the fact that the cells eventually differentiate tell us that the RNAi effect
lessens over time due to the promoter used or that the native RBR protein is
not needed after a certain developmental stage and thus the antisense RNA has
nothing to bind?

3)      Is
RBR a repressor; it seems like it is since the stem cells increase in a
loss-of-function experiment?

4)      They
discuss on pgs. 1338-9 that the number of cells increased in size is
significant, but is the actual cell size increase significant?

Not done already. Good experiment.

Good question. "Eventually differentiates" sounds like it overcomes a block (lack of RBR protein) but the proper controls were not included to make this conclusion.

It's thought to be a repressor of the cell cycle and a promoter of differentiation. Terminology can be confusing.

I couldn't find anything about increased cell size. Proximal meristem has increased #files of cells but still no significant increase in size of meristem.
