Questions for RBR Paper

Question 1) In the model on page 1345 the authors show a pathway for RBR pathway action and interaction in plants, however I feel that this model is a bit premature, is there any other evidence in plant systems to support this model?

Question 2) The authors have based the idea that the stem cells are maintained further away from the QC due to the action of RBR, however they state that there is a difference in the final size of the differentiated cells, could this effect be due to IAA maximum?

Question 3) I am not convinced that the RBR regulating the Stem cells is the entire story, how does IAA fit into the picture?

Authors do not discuss RBR transcript accumulation in plants. Other data on reducing RBR shows tissue-specific effects with altered cell sizes in epidermal cells of Arabidopsis. Model is premature (and animal-centric in my opinion).

2) The authors have based the idea that the stem cells are maintained further away from the QC due to the action of RBR, however they state that there is a difference in the final size of the differentiated cells, could this effect be due to IAA maximum?

Not sure about difference in final size - what figure?

3) I am not convinced that the RBR regulating the Stem cells is the entire story, how does IAA fit into the picture?

And, how can RBR potentiation of QC signal be that important if ablation of QC leads to rapid re-establishment of QC from "stem cells" or proximal meristem cells? Differentiation only reported in distal side of QC; stem cells on proximal side do not have a good marker. Marker is appearance of starch, so only columella side can be analzyed.
